Genome-Informed Multiplex Conventional PCR for Identification and Differentiation of <i>Xanthomonas citri</i> pv. <i>citri</i> Subpathotypes, the Causal Agents of Asiatic Citrus Canker

Asiatic citrus canker, one of the most important diseases citrus, is caused by Xanthomonas citri pv. citri. It has high economic impact and can spread easily, disease difficult to manage; it a quarantine organism in many citrus-producing countries. X. been separated into three subpathotypes (A, A * , w ) that differ host range geographical distribution, thus creating need differentiate for surveillance management. Availability useful diagnostic tools cornerstone successful surveillance, quarantine, eradication measures. In this study, multiplex conventional PCR (cPCR) assay was developed detection subpathotype determination Assay specificity assessed four different labs on total 146 58 other isolates. The demonstrated analytical sensitivity, specificity, selectivity. False negatives were observed with Lineage 2 strains, potential false positives bilvae. Combined simple extraction protocol, deployed successfully at Plant Protection Quarantine Pathogen Confirmatory Diagnostics Laboratory. This proven differentiating canker from symptomatic tissue.
